roaming | You can't perform calibrations in the desktop software. You can scale the data (for example, if you know all your load weights are 10% high, you can scale the data to compensate, but it's not the same as calibration). You can input manual values (moisture, weight, bu, etc.) for each load, if you know them, but it's a PITA and unbelievably time-consuming.
The best thing to do is to take your datacard back to the combine, calibrate in the combine, and then bring the updated data back to the desktop and archive it. This will update the desktop with the new calibrated data values...
